High-Yield Expression in E. Coli

Client Objective

A startup biopharmaceutical company currently in phase II with a proprietary therapeutic protein contracted with Vybion to develop a higher expressing clone to move into phase III. Current expression in yeast was inadequate to advance the product.

The Results

Vybion produced a codon and secondary-structure optimized gene, which was cloned into E. coli to develop extremely high-expressing clones.

The protein was purified from inclusion bodies with little loss.

The final recovery of purified protein was improved by nearly 40 fold over the recoveries obtained with yeast.

Vybion delivered the therapeutic protein, along with quality control documentation, fermentation, downstream processing and purification protocols for successful GMP manufacturing

The client contract with Vybion for a second product for which we developed expression and also developed this product in a pegylated format for animal PK/PD testing.
