The Protein Transport System
Vybion technology takes advantage of the twin arginine transport(TAT) system.
- Transports only folded proteins from the cytoplasm to the periplasmic space in E. coli
- Antibiotic reporter system is cloned 3' and used to select clones from a library of sequence changes
- Only properly folded proteins are transported and thus sensitive to antibiotic.
- Strength of antibiotic can also be adjusted to increase the stringency of selection criteria.
- Selection by growth on antibiotic is followed by colony PCR to identify specific sequence changes.

Antibody Selection Tools
- Accommodates more complex proteins than would otherwise be expressed in E. coli
- Single chain Fv selection enables identification of a larger, more diverse pool of high-affinity antibodies
- Proteins up to at least 140 kDa can be transported through the TAT pore
- Easily accommodates interacting polypeptides up to 45 kDa each comprising an scFv and antigen complex.
